The Blues were knocked out of the FA Cup after Poole-based side Hamworthy Recreation won 2-0 at the Camp & Satherley.
The newly promoted side will left off where they did last season, after winning Wessex Division One, with Max Wilcock grabbing both goals.
Kyle Critchell's side started the game well, earning a corner in the opening few minutes, however, Pat Jenkins spectacular bicycle kick went wide of the post.
In an exciting end to end start to the game at the Camp & Satherley, Hamworthy's first attack was defended well by George Stuttle.
With 16 minutes gone, Hamworthy created the first real chance of the game as Miles Wilcox connected with a corner but his header was cleared off the line by Jenkins.
With almost half an hour played, it would be the visitors who would take the lead. Tom Cooper whipped in a wonderful ball with Wilcock on hand to head past Harrison Bruton.
Hamworthy came close to doubling their soon after taking the lead but Tony Rolls header came crashing back off the crossbar.
As half-time approached, Hayden Scott hit a volley from range but it whistled just over the bar as Portland continued to push for an equaliser before the break.
Hamworthy's keeper Cal Watson was forced into a couple off late saves as the clock hit 45 minutes but the Blues were unable to find a way past the Rec keeper.
Hamworthy came out after the break all guns firing and almost instantly found a second, however Tyler Rudd's strike smashed the post bouncing out for a corner.
The Blues recovered well to control the opening 10 minutes of the half which resulted in Jenkins having an effort superbly saved by Watson.
With an hour on the clock, Jenkins was again involved after latching on to a ball over the top, but the forward's dangerous ball across the box was unable to find a teammate.
Just six minutes later and Hamworthy would double their lead with Wilcock on hand to head home at the back post from a deep cross.
As the game entered the final ten minutes there were calls for a penalty to the Blues for what looked like a deliberate handball in the box, but the ref was not interested.
The result means Portland drop out of the FA Cup and Hamworthy progress to the next round but Critchell and his team will now turn their attention to Tuesday's night's visit of Sherborne in the Wessex Premier season opener.
